Generation of anti-dextran monoclonal antibody and development of immunonephelometry for quantitative detection of dextran [Full subscriber]

The purpose of the study was to generate a monoclonal antibody against dextran and develop a method of immunonephelometry for quantitative detection of dextran. Dextran is covalently linked to carrier protein bovine serum albumin. An anti-dextran monoclonal antibody was obtained by hybridoma technology, which showed titer higher than 105 in mice ascites and the characteristics of the monoclonal antibody was studied. Then an immunonephelometric assay for quantitative detection of dextran was established, which showed a detection limit of 0.35 NTUs (nephelometric turbidity units) with a sensitivity of 5.5 μg/mL.
